I can watch Netflix on my laptop using my mobile hotspot. Will I have a problem trying to set up Roku so I can watch Netflix on my TV?

Should work fine. On your Roku network settings enter your hotspot SSID and password and you should be good to go. I use the Roku with an AT&T Mobley (4G) or, if the signal is strong enough, a Verizon mifi (3G).
