Colter Bay to West Yellowstone

Just go north on US191 and follow the signs to West Yellowstone.  I've done it in a 40' MH so you should be able to do it with your 5th wheel.  Go slowly in the park and you'll be fine.
 
X2 what Joel said. If you go early in the am you'll miss most of the park traffic.
 
191 to south entrance of Yellowstone and out West gate is the easiest, going west out of Jackson will send you over a pretty steep pass.

Not only is it a steep pass, it is narrow and very sharp turns.  SR-22 west out of Jackson is not suitable for large RV's.  Going South and west from Jackson is a really long way around.
